Ear Center: Digital Photography Using the OR Microscope
Camera Equipment
- Nikon D100 DSLR camera body (or similar body)
- 60mm F2.8 Micro Nikkor lens (This is a macro lens. The effective focal length is 90mm due to a 1.5x factor secondary to CCD size compared to 35mm film.)
Camera/Lens Settings
- Camera:
- Mode =Program
- ISO = 400
- White Balance = incandescent light
- File Size = medium jpeg (if you are using the images for PowerPoint slides)
- Lens : Set lens to "manual focus" (adjust the ring on the lens barrel) and adjust the focus to "infinity" on the distance scale
Technique
- Adjust camera body and lens settings as above
- Accurately focus the microscope on the field
- Hold the lens against the microscope's rubber cup on the ocular: compose the image & shoot
- check the image for content and focus
Post-Processing of the Image
Post-process in Photoshop CS4 or similar software program (Photoshop Elements, etc.) Perform the following in order:
- Crop the image appropriately, then set image size to 150 pixels
- Adjust Levels, Contrast, and Color
- Adjust Brightness and Contrast
- Save the image as a jpeg file in an appropriate folder
- Always make backup copies
